For high throughput screening of HIF-1α-targeted drug, we need to determine the activity sensitively and quantitatively. In the present study, we proposed a method of fluorescence correlation analysis on HIF-1α activation in which Co2+ treatment against stable transformants of GFP-labelled HIF-1α mimicked hypoxia. In fluorescence correlation spectroscopy, we observed fluorescence intensity fluctuation within a volume element which fluorescence molecules enter and exit because of the Brownian motion in solution or cytosol. When one-component model was used for the analysis, it was difficult to discriminate diffusion coefficient of active form of HIF-1α with inactive one. In two-component model, however, a fraction of slow moving component, GFP-labelled HIF-1α increased significantly when the transformants were exposed to Co2+. In the case of high throughput screening for HIF-1α-targeted drug with fluorescence correlation spectroscopy, we should use the fraction of the slower moving component to judge the activation.","subitem_description_type":"Abstract"}]},"item_creator":{"attribute_name":"著者","attribute_type":"creator","attribute_value_mlt":[{"creatorNames":[{"creatorName":"田中, 悠樹"}],"nameIdentifiers":[{}]},{"creatorNames":[{"creatorName":"野村, 保友"}],"nameIdentifiers":[{}]},{"creatorNames":[{"creatorName":"タナカ, ユウキ"}],"nameIdentifiers":[{}]},{"creatorNames":[{"creatorName":"ノムラ, ヤストモ"}],"nameIdentifiers":[{}]},{"creatorNames":[{"creatorName":"Tanaka, Yuki"}],"nameIdentifiers":[{}]},{"creatorNames":[{"creatorName":"Nomura, Yasutomo"}],"nameIdentifiers":[{}]}]},"item_files":{"attribute_name":"ファイル情報","attribute_type":"file","attribute_value_mlt":[{"accessrole":"open_access","displaytype":"detail","filename":"05論文(野村)(修正後).pdf","filesize":[{"value":"395.6 kB"}],"format":"application/pdf","licensetype":"license_note","mimetype":"application/pdf","url":{"url":"https://maebashi-it.repo.nii.ac.jp/record/21/files/05論文(野村)(修正後).pdf"},"version_id":"2c1f20f3-9f00-4e01-8d86-1bbfed14becb"}]},"item_keyword":{"attribute_name":"キーワード","attribute_value_mlt":[{"subitem_subject":"低酸素誘導因子"},{"subitem_subject":"蛍光相関分光法"},{"subitem_subject":"ブラウン運動"},{"subitem_subject":"拡散方程式"},{"subitem_subject":"拡散定数"},{"subitem_subject":"HIF-1α"},{"subitem_subject":"Fluorescence correlation spectroscopy"},{"subitem_subject":"Brownian motion"},{"subitem_subject":"Diffusion equation"},{"subitem_subject":"Diffusion coefficient"}]},"item_language":{"attribute_name":"言語","attribute_value_mlt":[{"subitem_language":"jpn"}]},"item_title":"蛍光性低酸素誘導因子の相関分析","item_titles":{"attribute_name":"タイトル","attribute_value_mlt":[{"subitem_title":"蛍光性低酸素誘導因子の相関分析"}]},"item_type_id":"3","owner":"2","path":["10"],"pubdate":{"attribute_name":"公開日","attribute_value":"2015-11-10"},"publish_date":"2015-11-10","publish_status":"0","recid":"21","relation_version_is_last":true,"title":["蛍光性低酸素誘導因子の相関分析"],"weko_creator_id":"2","weko_shared_id":2},"updated":"2023-06-20T14:16:42.232075+00:00"}
